A simplified procedure for purification of large amounts of poliovirus: characterization and amino acid analysis of Type 1 poliovirus.

A sign&ant advance in the study of animal viruses was marked by the work of Schwerdt and Schaffer (l), who first obtamed purified poliovirus in quantities sufficient to permit rigorous chemical and physical characterization. Their procedure, however, required large volumes of starting material, and was both lengthy and laborious. The development of suspension culture techniques (2) has provided a convenient means for growing mammalian cells under conditions which permit the controlled production of relatively large amounts of poliovirus (3). Such infected cultures have been utilized as the starting material for a rapid and convenient purification procedure which embodies the chromatographic method of Hoyer et al. (4, 5). The present report describes the application of this procedure to the purification of Type 1 poliovirus, and the chemical, physical, and immunological characterization of the purified virus. The accompanying paper (6) deals with a study of the origin and synthesis of the viral protein.
